Speculation started after Disney released a clip of the new film, which is due out June 16.
In one scene Tank the octopus gets into a baby's buggy and two women turn around shocked their baby has transformed.
One viewer suggested the women were a couple and it sent Twitter into overdrive.
While many were full of praise if the rumour is indeed true, some declared they would be boycotting the film.
And others said the speculation was premature as it could just be "two women standing next to each other".
Others pointed out that Ellen DeGeneres - one of Hollywood's most famous lesbians - voices Dory.
And others seemed to be a little confused by the rumour...
The sequel to 2003's Finding Nemo - which was hugely popular and grossed more than $380 million at box office - centres on Dory's quest to find her parents.
Dory is helped along the way by Nemo and his father Marlin.
Diane Keaton, Idris Elba, and Michael Sheen will also be popping up at some point in Dory's journey, according to reports.
